Obesity care is moving quickly past the one-drug, one-pathway era. Current therapies have reshaped what treatment can achieve, yet responses vary widely and not every patient tolerates or responds to them, leaving clinicians searching for the next complementary mechanism. Amylin is among the most compelling. A hormone most clinicians last encountered as insulin’s co‑secreted partner, amylin is now understood as a systems‑level satiety signal with distinctive receptor biology and promise as monotherapy or in combination strategies. For many obesity‑medicine clinicians, though, that knowledge still stops at “co‑secreted with insulin,” a gap that makes it hard to interpret the growing body of investigational amylin data, distinguish selective amylin analogs from dual amylin and calcitonin receptor agonists (DACRAs), or judge where these agents fit within chronic, multi‑pathway disease management.
In this second CMEO Snack of the Riding the Waves of Obesity Science series, amylin researcher Matthew R. Hayes, PhD joins moderator Robert F. Kushner, MD for a focused, practical conversation that turns amylin from a half‑remembered footnote into working clinical knowledge; what amylin actually does, how its calcitonin‑receptor/receptor activity-modifying protein (RAMP) biology shapes today’s drug classes, and why its signal complements rather than duplicates glucagon-like peptide-1 (GLP‑1).
